چکیده

Due to the biological importance of long chain fatty acyl-CoA synthetase (ACSL), ACSL has been studied extensively in the past quarter century. ACSL is a ubiquitous enzyme from bacteria to mammal, and its product, long chain fatty acyl-CoA, is well known to be utilized for the energy metabolism. Furthermore ACSL participates in the cellular uptake of the exogenous Correspondence/Reprint request: Dr. Hideo Ago, Structural Biophysics Laboratory, RIKEN SPring-8 Center Harima Institute, 1-1-1Kouto, Sayo, Hyogo 679-5148, Japan. The unique catalytic mechanism, by which the fatty acyl-CoA as the final product is formed unidirectionally via the stable intermediate fatty acylAMP, is behind the biological functions of ACSL. In this manuscript, the unique catalytic mechanism is reviewed with respect to 3-dimentional structure of ACSL.

The Mycobacterium tuberculosis very-long-chain fatty acyl-CoA synthetase: structural basis for housing lipid substrates longer than the enzyme.

The Mycobacterium tuberculosis acid-induced operon MymA encodes the fatty acyl-CoA synthetase FadD13 and is essential for virulence and intracellular growth of the pathogen. Fatty acyl-CoA synthetases activate lipids before entering into the metabolic pathways and are also involved in transmembrane lipid transport. Unlike soluble fatty acyl-CoA synthetases, but like the mammalian integral-membr...

Biochemical demonstration of the involvement of fatty acyl-CoA synthetase in fatty acid translocation across the plasma membrane.

Fatty acyl-CoA synthetase, the first enzyme of the beta-oxidation pathway, has been proposed to be involved in long chain fatty acid translocation across the plasma membrane of prokaryotic and eukaryotic cells. To test this proposal, we used an in vitro system consisting of Escherichia coli inner (plasma) membrane vesicles containing differing amounts of trapped fatty acyl-CoA synthetase and it...

The effects of ginsenoside Rb1 on fatty acid β-oxidation, mediated by AMPK, in the failing heart

Objective(s): This study intended to investigate the effects of Ginsenoside-Rbl (Gs-Rbl) on fatty acid β-oxidation (FAO) in rat failing heart and to identify potential mechanisms of Gs-Rbl improving heart failure (HF) by FAO pathway dependent on AMP-activated protein kinase (AMPK). Materials and Methods: Rats with chronic HF, induced by adriamycin (Adr), were randomly grouped into 7 groups. Gs-...

Transmembrane movement of exogenous long-chain fatty acids: proteins, enzymes, and vectorial esterification.

The processes that govern the regulated transport of long-chain fatty acids across the plasma membrane are quite distinct compared to counterparts involved in the transport of hydrophilic solutes such as sugars and amino acids. These differences stem from the unique physical and chemical properties of long-chain fatty acids. To date, several distinct classes of proteins have been shown to parti...

Human fatty acid transport protein 2a/very long chain acyl-CoA synthetase 1 (FATP2a/Acsvl1) has a preference in mediating the channeling of exogenous n-3 fatty acids into phosphatidylinositol.

The trafficking of fatty acids across the membrane and into downstream metabolic pathways requires their activation to CoA thioesters. Members of the fatty acid transport protein/very long chain acyl-CoA synthetase (FATP/Acsvl) family are emerging as key players in the trafficking of exogenous fatty acids into the cell and in intracellular fatty acid homeostasis. We have expressed two naturally...
